Several police cars were called to Club X in Queen Street about 8.15pm.
Police confirmed Thursday morning the person threatened staff with a knife before fleeing the store with a quantity of cash.
Police reported no injuries.
Detective Michael Thompson described the offender as male and wearing dark blue tracksuit pants and a dark blue hoodie.
The offender fled across Queen Street and up Lyttleton Terrace to Mitchell Street.
"He ran past witnesses so we are keen to talk to anyone who might have information," Detective Thompson said.
Police searched the CBD for the offender.
Meanwhile three patrol cars descended on the Bendigo Railway Station about 8.45pm Wednesday.
The Bendigo Advertiser is unsure whether this incident is related to the hold up at Club X.
