British bands The Kooks and Example lead the international acts while Tame Impala, The Temper Trap and Regurgitator will feature as some of the Australian acts.
Canadian duo Tegan and Sara are set to return after a successful tour in 2010, while up-and-coming bands Flume, Alpine, Last Dinosaurs and Hungry Kids of Hungary are also confirmed to be playing.
More than 75,000 people are expected to attend the five shows on the regional tour.
Last year, the Bendigo leg of the festival sold out in less than a week.
“After six months of planning, it’s always an exciting time when the GTM line up is finally revealed," GTM promoter Stephen Halpin said.
"Again we've assembled a great bunch of exciting new acts and festival favourites.
"We’re very excited to be hosting They Might Be Giants' first visit to Australia in a long while."
The GTM tour arrives at Bendigo's Prince of Wales Showgrounds on Saturday, May 4.
Tickets for the festival are $99.90 and go on sale for the Bendigo leg on Wednesday, February 6.
